The Applied Biosystems OpenArray AccuFill System is an automated, high-throughput sample loading platform designed to precisely transfer DNA samples and PCR master mix from standard 384-well plates onto OpenArray Plates for real-time PCR analysis. The system uses capillary action to draw samples into AccuFill tips and dispense them evenly across the OpenArray plate surface, where self-metering through-holes automatically fill to the correct reaction volume without the need for pressure-based dispensing or manual volume control. Because the OpenArray format contains no closed wells, air bubbles are not trapped during loading, resulting in highly uniform fills and excellent data consistency.
Optimized specifically for downstream real-time PCR workflows, the AccuFill System supports SYBR, OpenArray Real-Time PCR, TaqMan OpenArray Real-Time PCR, and TaqMan OpenArray Genotyping chemistries. OpenArray technology significantly increases throughput and efficiency: a single QuantStudio™ 12K Flex OpenArray plate is equivalent to eight conventional 384-well plates, and the system can process up to four OpenArray plates per run. This enables laboratories to analyze up to 1,728 genotyping samples or 2,304 gene expression samples within an 8-hour workday, with the option for offline thermal cycling to further optimize instrument utilization.
The accompanying OpenArray AccuFill v2.0 software provides end-to-end workflow control, including sample plate setup, mapping from 96-well to 384-well formats, support for multi-sample loading, and generation of all required plate files (TPF, SPF, loaded files, and CSV outputs) for compatible Applied Biosystems™ real-time PCR instruments. The software also includes plate orientation correction, loading history tracking, and sample integration capabilities to streamline data management. Designed for reliability and reproducibility, the system delivers a typical fill rate of 99.25%, with maximum allowable sample and assay carryover below 1% and less than 5% data loss due to evaporation, making it a robust solution for high-density, high-confidence qPCR applications
